You really mean “how can I know if 3d card supports T&L?” OR
You actually mean “how can my software know if 3d card supports T&L?”

In the first case, you can look at the spec of the graphics card.

What actually is that T&L?
Is it just a hardware acceleration for transform and lighting (done with openGl) or do I have to do something to get my program use it?

if your graphics card does hardware t&l there is nothing special you have to do to use it, although there are some techniques which use it better than others (i think nvidias developer page has more info on this subject)
